Highly specific PTM antibodies should meet the following two conditions:
Target Protein Specificity Verification:
Reducing the expression level of the target protein via knockdown (KD) or knockout (KO) should result in a significant reduction or complete loss of the antibody detection signal, confirming high specificity toward the target protein.
Modification Site Specificity Verification:
Mutating the amino acid at the modification site to a non-modifiable residue (e.g., alanine substitution) should lead to a complete loss of antibody signal, demonstrating that the antibody specifically recognizes the post-translational modification at that site and thus verifying its site specificity.
